This event will take place in Smilow 9-146 and on zoom from 4PM – 5PM. \n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\nDespite active cell proliferation\, differentiation and apoptosis\, mammalian organs maintain their size and function\, indicative of a delicately balanced program that regulates cell population dynamics. Dysregulation of this homeostasis results in imbalances that can give rise to aging\, and aging-related diseases(e.g.\, neurodegenerative disorders and cancer). The central vision of the Cao Lab is to investigate how does a cell population maintain homeostasis and how it is disrupted in aging and diseases. Our primary approach is to develop state-of-the-art genomic techniques to profile and perturb cell dynamics at single-cell resolution and dissecting its connection with internal transcriptome state and external cell-cell interactions. Our long-term goal is to leverage this knowledge to develop cell-type-specific engineering tools to enhance cell population robustness or restore cell population homeostasis. \n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n

However\, these large multi-subunit complexes also contain many other proteins of unknown function that are critical for development and highly mutated in human disease. Our goal is to use a combination of biochemical and epigenomic assays to understand the function of these complexes during normal cellular differentiation and the mechanistic basis for diseases caused by mutations in BAF complex subunits. We are focusing on cellular models of inflammation\, cancer\, and pluripotency. \n\n\n\n\n\n\n\n\n\n

The lab is currently focused on understanding how early life stress increases sensitivity to subsequent stress. This research is translationally motivated by the robust clinical finding that child maltreatment and other forms of early life stress increase the lifetime risk of depression and other mood\, anxiety\, and drug disorders. Studies in humans and animals suggest that early life stress sensitizes individuals to stress later in life\, leading to a first appearance or synergistic worsening of depression-like symptoms after additional stress.

Dr. Kueh will be giving a talk titled\, “Understanding and engineering immune cell fate control circuits.” This event will take place in Smilow 10-146 and on zoom from 4PM – 5PM. \nThe immune system protects the body against a wide array of threats\, ranging from viruses to bacteria to cancer.  These versatile defense capabilities are upheld by a collection of immune cell types\, each of which can sense and mount responses tailored towards distinct types of threats. \nThe Kueh Lab seeks to elucidate how immune cells fate decisions\, both to develop from stem cells\, and to mount effective responses to challenges.  Immune cells utilize circuits of interacting genes and proteins to sense signals and execute cell state transitions.  To understand how these signaling and gene regulatory circuits work\, we use powerful reporter systems and quantitative long-term imaging to follow circuit activity in single cells.  We also use cutting-edge single-cell genomics and genome engineering techniques to define their molecular parts and architecture\, and use mathematical modeling to test and predict their functional performance.  Through this work\, we aim to lay foundations to rationally program immune cells to fight cancer and other life threatening diseases.

DESCRIPTION:Lamia Wahba\, Ph.D.\nAssistant Professor\nThe Rockefeller University\nThe Wahba Lab \nDr. Wahba did her graduate work at the Carnegie Institute’s Dept of Embryology\, located on the Hopkins campus. There she worked on R-loops in budding yeast under the mentorship of Douglas Koshland. For her postdoc she moved to Andy Fire’s lab at Stanford to study small RNA systems in the free-living nematode Caenorhabditis elegans. She joined Rockefeller as an assistant professor in 2023.      \nThe Wahba lab seeks to understand the role of nongenetic inheritance mechanisms in disease and evolutionary processes by dissecting the processes and parameters influencing it\, the biological functions impacted\, and their physiological relevance. Research in the lab builds on Wahba’s previous findings about the role of small RNAs and their protein partners\, Argonautes\, in the transgenerational maintenance of the germline’s immortal character. In most metazoans\, Argonaute/sRNA systems are central to the transgenerational inheritance of gene regulation programs\, but a full mechanistic understanding of initiation and inheritance determinants remains under investigation. \nUsing the free-living nematode C. elegans as a model system\, Wahba’s work has found that some heritable sRNAs propagate through self-perpetuating amplification cycles\, a strategy that allows for the indefinite spread of silencing signals across generations but risks overamplifying them. To limit the potentially dire consequences of sRNA overamplification\, Wahba identified the piRNA pathway (involving a second type of Argonaute/sRNA ribonucleoprotein) as an essential negative modulator of it. In the absence of piRNAs\, worms progressively lose fertility over multiple generations until they become completely sterile\, the result of genes targeted by endogenous Argonautes/sRNAs succumbing to perpetual and inappropriate transgenerational silencing. \nThe roles Argonautes/sRNAs play in maintaining germline mortality highlight the potent nature of nongenetic inheritance\, and the existence of molecular machineries dedicated to its active limitation and regulation. By delineating the mechanistic principles underlying acquisition and inheritance of nongenetic information\, the Wahba lab’s ultimate hope is to address a fundamental question facing researchers across many biological fields: whether it is time to revamp the framework for inheritance to incorporate transmission of molecular factors that transcend DNA sequences. \nHosted by Colin Conine\, Ph.D.

DESCRIPTION:Lamia Wahba\, Ph.D.\nAssistant Professor\nThe Rockefeller University\nThe Wahba Lab \nDr. Wahba did her graduate work at the Carnegie Institute’s Dept of Embryology\, located on the Hopkins campus. There she worked on R-loops in budding yeast under the mentorship of Douglas Koshland. For her postdoc she moved to Andy Fire’s lab at Stanford to study small RNA systems in the free-living nematode Caenorhabditis elegans. She joined Rockefeller as an assistant professor in 2023.      \nThe Wahba lab seeks to understand the role of nongenetic inheritance mechanisms in disease and evolutionary processes by dissecting the processes and parameters influencing it\, the biological functions impacted\, and their physiological relevance. Research in the lab builds on Wahba’s previous findings about the role of small RNAs and their protein partners\, Argonautes\, in the transgenerational maintenance of the germline’s immortal character. In most metazoans\, Argonaute/sRNA systems are central to the transgenerational inheritance of gene regulation programs\, but a full mechanistic understanding of initiation and inheritance determinants remains under investigation. \nUsing the free-living nematode C. elegans as a model system\, Wahba’s work has found that some heritable sRNAs propagate through self-perpetuating amplification cycles\, a strategy that allows for the indefinite spread of silencing signals across generations but risks overamplifying them. To limit the potentially dire consequences of sRNA overamplification\, Wahba identified the piRNA pathway (involving a second type of Argonaute/sRNA ribonucleoprotein) as an essential negative modulator of it. In the absence of piRNAs\, worms progressively lose fertility over multiple generations until they become completely sterile\, the result of genes targeted by endogenous Argonautes/sRNAs succumbing to perpetual and inappropriate transgenerational silencing. \nThe roles Argonautes/sRNAs play in maintaining germline mortality highlight the potent nature of nongenetic inheritance\, and the existence of molecular machineries dedicated to its active limitation and regulation. By delineating the mechanistic principles underlying acquisition and inheritance of nongenetic information\, the Wahba lab’s ultimate hope is to address a fundamental question facing researchers across many biological fields: whether it is time to revamp the framework for inheritance to incorporate transmission of molecular factors that transcend DNA sequences. \nHosted by Colin Conine\, Ph.D.
